I'd like to create a website where people can click on certain dates in an interactive calendar, signaling when they'll be absent from school. Is there an API that exists that could provide functionality like highlighting selected dates in green after they've been clicked on?

Check out jQuery UI's datepicker and Google's calendar API. You could create a form that lets people create appointments that save to a google calendar you own.

Just a side note. APIs are generally used to store and retrieve data (like the actual appointment name, start date and end date). Perhaps you are looking for a front-end library or jQuery plugin to render a calendar
